Welcome to the PCHPI - Pittsburgh Center for HIV Protein Interactions

 

Elucidating the interactions of human immunodeficiency virus (HIV) proteins with those of the host cell is crucial for understanding viral replication and pathogenesis. Knowledge of these processes, including the spatial and temporal details of the intricately interwoven choreography between pathogen and host components, will provide an essential foundation for developing focused and effective preventative and therapeutic strategies in the fight against AIDS. Although large numbers of putative interactions have been reported in the scientific literature, structural confirmation of these interactions and/or reliable assessments about their functional importance are difficult to ascertain.


We have established an interdisciplinary Center for HIV Protein Interactions dedicated to determining high-resolution structures of HIV proteins and their interacting partners in the human host. The Center seeks to define events, pathways, and host cell factors involved in HIV cellular infection, prior to integration of the viral genome into that of the infected cell, and aims to characterize these structurally by X-ray crystallography, NMR, and cryo Electron Microscopy. Results will provide insights into detailed aspects of HIV immediate post-entry biology and it is anticipated that they can be effectively leveraged for developing novel therapeutic strategies.
